Mode Of Action

Reserpine is a pure crystalline alkaloid of Rauwolfia. Reserpine acts by depleting stores of catecholamines and 5-hydroxytryptamine in many organs, resulting in a decreased heart rate and a lowering of arterial blood pressure. Clopamide is a thiazide diuretic. Dihydroergocristine is dihydrogenated ergot alkaloids whose mechanism of action is not fully understood. It is thought to act centrally to decrease vascular tone and slows the heart rate, and acts peripherally to block alpha-receptors.
Drug Indication Essential hypertension; arterial and renal hypertension.
Precautions Impaired hepatic and/or renal functions; diabetes; gout; monitored serum potassium and uric acid levels; epilepsy; concurrent administration with levodopa; cardiac infarction; cardiac damage.
Contra-Indications Sulphonamide sensitivity; persistent hypokalaemia; depression; acute gastric/duodenal ulcer; pregnancy; lactation; ulcerative colitis; direct acting sympathomimetics.
Side Effects Postural hypotension; blood dyscrasia; headache; GI disturbances; flushing; depression; suicidal tendency; nasal congestion; glycosuria in diabetes; gout; muscle spasm; dry mouth; parkinson-like syndrome; increased appetite; loss of libido.
